T-Lymphotropic and Neurotropic Herpesviruses

Research Summary

Our laboratory is interested in the host-cell interaction of lymphotropic and neurotropic herpesviruses and their pathogenic functions.

1. Rhadinoviral T-cell vectors

The T-lymphotropic rhadinovirus herpesvirus saimiri (HVS) transforms human T cells to stable growth in culture, which then proliferate independently of antigen or mitogen and retain the antigen-specificity and many other features of their parental clones. The virus is not released from the transformed cells and persists in form of non-integrated episomes. These features of HVS are promising for the amplification of functional T cells and for gene transfer and functional expression of foreign genes in this cell type. We apply this episomal gene transfer system for instance for the redirection of T-cellular antigen specificity towards tumor antigens in experimental immunotherapy.


2. Cytokines of the interleukin-10 Family

By subtractive hybridization, we cloned a cellular gene (ak155) with 25% identity and structural similarity to interleukin-10 (IL-10), from HVS-transformed human T-cells. Specifically, these cells overexpress ak155 and secrete the derived protein into the supernatant. In other T-cell lines and in native peripheral blood cells, ak155 is transcribed at low level. The protein forms homodimers similarly to IL-10 and induces STAT phosphorylation in epithelial target cells via members of the type-2 family of cytokine receptors. AK155, also termed IL-26, belongs to a newly described family of cytokines which are structurally, but not functionally, related to IL-10.

3. Neuropathy by Varicella-Zoster virus

The neurotropic human alpha-2 herpesvirus Varicella-Zoster virus (VZV) causes chickenpox during primary infection. The reactivation of VZV from its persistent state in sensory neurons induces shingles (Herpes zoster) which are characterized by severe, long-lasting pain and often lead to postherpetic neuralgia. In cooperation with Prof. Michaela Kress (Institute for Physiology, Innsbruck) we described a virus infection-related induction of noradrenaline sensitivity of sensory neurons in culture. By using recombineering techniques, we are studying recombinant herpesviruses in order to further investigate the mechanism of viral pain induction.
